On March 22, local time, the second round of the men's singles at the 2025 ATP1000 Miami Masters began, and Chinese player Buyun Chaokert lost 4-6/4-6 to the tournament's No. 10 seed De Minaur in the second round, since last year's US Open, Buyun Chaokert has won 1 win and 6 losses in seven matches against the TOP10 (defeated Rublev in the 1/4 finals of the 2024 China Open).

Hong Kong's Wong Chak Lam, a wildcard player, won 7-6(3), 2-6, 7-6(5) against local favourite and 13th seed Shelton 7-6(3), 2-6, 7-6(5) to win the first career match against a top-20 player and advance to the third round of the Masters against lucky loser Walton.
Top seed Zverev defeated Great Britain's Fearnley 6-2, 6-4 to set up a third-round clash with Thompson. Homegrown No. 1 and third seed Fritz eliminated Sonego 7-6(2), 6-3 to face Shapovalov in the third round. Draper and Rublev were eliminated in the second round, with Czech rookie Mensik beating Draper, who won his first Masters title at Indian Wells last week, 7-6(2), 7-6(3), to face Safiulin in the third round. Eighth seed Rublev lost 5-7, 4-6 to Berges and suffered back-to-back rounds in the Sunshine Doubles. Indian Wells runner-up and 11th seed Rune were beaten 6-4, 3-6, 6-7(5) by homegrown cannon Opelka in the second round.
